Presentation will first focus on studies in which sequence changes were introduced in the cytoplasmic tail of HIV-1 gp41. These immunogens were evaluated in vitro and in vivo. The objective was to yield a more stable envelope on the cell surface; thereby, increasing the longevity of protein spikes on the surface, exposure of masked epitopes and antibody responses of greater magnitude and breadth.
Second focus of the presentation will be on adjuvants and immunomodulatory agents to augment the immune response following HIV-1 vaccination. For this, we will evaluate mucosal and systemic immune responses in mice using two novel inulin-based adjuvant formulations and by using an E3 ligase.
